Establishment and Characterization of Two New Brain Tumor Cell Lines

Abstract

|
|
Objective: This study established cell lines from surgical specimens removed from brain tumor cases. Explant cultures of fresh tumor tissue were used to develop new permanant cell lines of anaplastic astrocytoma(Pusan National University Neurosurgery Glioma-13, PNUNS G-13) and atypical meningioma(PNUNS Meningioma-21, PNUNS M-21).
Methods: The morphology and ultrastructure of these cell lines were checked by electron microscope. Karyotyping was performed by in situ coverslip culture method. Tumorigenesis and growth patterns of tumors from these cell lines were confirmed by xenograft study of implantation to the nude mice.
Results: The results obtained were the followings. 1) Cell line£ºPNUNS G-13 and PNUNS M-21 in its 30th passage were established. 2) Double time£ºDouble times of PNUHNS G-13 and M-21 were 60 and 54 hours respectively. 3) Electron microscopic examination£ºPNUNS G-13 showed irregular nuclei and cytoplasm contained vesicles of distended rough endoplasmic reticulum, glial fibrillary acidic protein(GFAP) and internal filament. PNUNS M-21 showed irregular large nuclei and the cytoplasm dominated by numerous filaments. 4) Karyotype analysis£ºIn PNUNS G-13, the chromosome number and structure showed normal karyotype(46, XX). But in PNUNS M-21, it showed multiple numerical and structural abnormalities. The abnormal clonality was 92, XXXX, del(2)(pter ¡æ cen ¡æ q33;), del(16)(pter ¡æ cen ¡æ q22;). 5) Xenograft study£ºPNUNS G-13 produced progressively growing tumors within 2-4weeks. PNUNS M-21 had no exact tumor growing. Histologically, tumors of nude mice were similar to the original tumor.
Conclusion: The establishment and characterization of the cell line might accomplish the basic study in mechanism of tumorigenesis and treatment of tumors.
